The two- and three-dimensional visualization and mapping of fibre tracts in the human brain is a highly difficult task. Effective brain mapping does not only serve basic research in Neuroanatomy, but does also have a significant relevance in treatment of complex diseases. Large fibre tracts can already be seen by means of macroscopic techniques like the dissection technique developed by Klingler. There are even more techniques, all with certain advantages and disadvantages. All methods are lacking neuroanatomic validation and so far there is no standard-method.

One physical feature of the myelin has not been used up to now: the birefringence. The myelin is able to turn polarized light in relation to the direction and incline of the corresponding fibre. After passing an analyzer and digital processing one is able to display the two- and three-dimensional course of fibre-tracts. Not only large fibre tracts (e.g. Corpus callosum, internal capsule) could be shown, but also smaller tracts as for example Fibrae arcuatae breves.

The method described in this book is the groundwork for now further developed methods like PLI (Polarized Light Imaging).
